New methods for neutrophil identification and diagnosis of acute lymphoblastic leukemia with KMT2A rearrangement
The offer refers to 2 inventions created at Jagiellonian University.
1. First invention relates to a new and innovative method of confirming the presence of rearrangements within the KMT2A gene in acute lymphoblastic leukemia (ALL) cells treated with an aldehyde fixative, which consists measuring of the integral intensity ratio of the two bands present in the Raman spectra of blasts collected from blood or bone marrow of patients diagnosed towards ALL and fixed with an aldehyde solution.
2. The second invention relates to a new and innovative method of confirming the differentiation towards neutrophils of human promyelocytic leukemia cells of the HL-60 line, which consists of calculating the ratio of the intensities of Raman bands characteristic for cytochrome, lipids and the MitoBADY reporter in the Raman spectra of living cells.
The aim of the inventions is to develop a new, rapid and molecular-oriented method of identifying the differentiation of human promyelocytic leukaemia cells of the HL-60 line and/or the presence of rearrangements within the KMT2A gene in ALL cells.
These techniques are met by Raman spectroscopy, which is a fast and non-destructive method for the tested sample. It allows one to obtain comprehensive biochemical information from cells without the need to use any tracers. MitoBADY, which is a Raman reporter enhance the sensitivity and specificity of the developed method. The proposed inventions postulate the use of Raman spectroscopy, commonly used in quality control, and simple chemometric analysis (which can be successfully automated) as an approach that will enable precise and unambiguous identification of the high-risk molecular subtype of leukaemia or neutrophil-like cells.
