Description of the project area: Antibiotic resistance in bacteria, including Acinetobacter baumannii, represents a critical challenge in global healthcare. Understanding the molecular mechanisms underlying bacterial RNA metabolism is essential for developing novel therapeutic strategies. This project focuses on characterizing the protein interactions involving RNase E, a key enzyme in RNA processing and degradation in A. baumannii. RNase E in E. coli forms a multicomponent complex, known as the RNA degradosome. By identifying RNase E interacting proteins through affinity purification and mass spectrometry, this project aims to elucidate the composition and functional dynamics of the RNase E complex, offering potential targets for antibacterial drug development.
Job description: The PhD student will utilize techniques such as affinity purification and mass spectrometry to identify components of the RNA degradosome in Acinetobacter baumannii. Additionally, the student will employ biochemical methods and single-molecule microscopy to investigate the functional roles of these proteins in RNA metabolism. This position integrates microbiology, molecular biology, and biochemistry, and seeks a candidate with a strong interest in RNA biology and antibiotic resistance.
Laboratory interest: The Laboratory of Single-Molecule Biophysics is dedicated to investigating the regulation of gene expression in bacteria through in vivo and in vitro methods, such as cutting-edge single-molecule TIRF microscopy. Another research branch within the laboratory is focused on exploring host-bacteriophage relationships.
