Program wykładów:
1. Fundamentals of CRISPR-Cas9 Gene Editing
- Introduction - elementary concepts of how CRISPR-Cas9 gene editing works, the basic reagents needed, and the basic workflow of a gene editing experiment using CRISPR-Cas9 products.
- Highlights of the primary applications for which this technology is currently being used.
2. A comparison of RNAi and gene editing
- Considerations in choosing between using gene editing or RNAi.
- Review of reagents available for gene silencing applications (siRNA and shRNA tools) and how to choose which ones will best meet your experimental needs.
- Introduction of Accell siRNA for difficult to transfect cells.
3. Considerations for a CRISPR-Cas9 experiment
- Considerations for designing a gene editing experiment including how to choose which Cas9 format is best for your experiment, and selecting between a crRNA and sgRNA approach.
- Bioinformatic considerations involved in designing these tools.
- Importance of controls and the “dos and don’ts” to consider when designing and troubleshooting your experiments.
